IpIBA seminar series 2022/2023 “Conjugation of nanoantibodies with NIR fluorophores as molecular probes for fluorescence-guided surgery”
Fluorescence-guided surgery (FGS) is an imaging technique that allows visualization of structures and tissues that may not be distinguishable under white light, providing real-time information to the surgeon. FGS is increasingly linked to molecular imaging. It uses probes that provide information on physiological and pathological processes, such as antibodies, peptides or small molecules. A very innovative alternative is the use of nanoantibodies (Nanobody, Nb), consisting of single domain variable regions (VHHs) derived from single heavy chain antibodies present in camelids. Nbs present several advantages for FGS in vivo including their small size (~15kDa) which favors greater tissue penetration and faster clearance than other antibodies. There is a wide variety of fluorescent molecules that can be used for Nbs-mediated FGS.
Among them, fluorophores emitting in the near infrared (NIR), where tissue autofluorescence interference is minimal, stand out. In our laboratory we have developed and characterized Nbs against different tumoral biomarkers. We are evaluating different conjugation strategies with NIR fluorophores (IRDye800CW), since the chosen strategy significantly influences the properties of the nanobody such as its biodistribution and the contrast it provides on adjacent tissue.
